Introduction: Recent studies show that feeding Atlantic cod (Gadus morhua) with wild zooplankton, as opposed to enriched rotifers, can provide a better scope for growth during the larval period, as well as have long-term beneficial effects on juveniles (Imsland et al., 2006, Koedijk et al., 2010). Other studies have shown improved growth and survival in fish larvae fed fish protein hydrolysate as part of their diet (Cahu et al. 1999., Zambonini-Infante et al. 1997). However, the factor(s) mediating this enhanced growth have not been identified. This research had two objectives: 1) Conduct trials in Newfoundland to examine whether partial dietary supplementation with zooplankton (5 10% of total prey items) and fish protein hydrolysate show similar results with regards to cod growth and production traits. 2) Use a multi-faceted approach to identify what factor(s) is/are mediating the enhanced growth, survival etc.
Rearing: Eggs from communally spawned Atlantic incubated at 6-7 o C until 100% hatch. Cod larvae stocked in 400 L tanks at a density of 50 larvae / liter and randomly divided into 3 different treatments, based on feeding regime/diet (see Fig. 1) : A) RA-O: Rotifers / Artemia, enriched with Ori-Green (6 replicates). B) RA-PH: Ori-Green enriched Rotifers / Artemia and Protein Hydrolysate enriched Rotifers / Artemia (3 times a week)(6 replicates). PH purchased from IceProtein Ltd. (Iceland) C) RA-Zoo: Ori-Green enriched Rotifers/Artemia with 5-10% supplementation with wild zooplankton (Tempora, Oithona, Pseudocalanus) (4 replicates). Potters clay used to increase tank turbidity and reduce bacterial numbers, photoperiod 24 h light, temperature 10 o C. Zooplankton collected from Conception Bay (Newfoundland) using a 100 µm mesh plankton net with 1 m diameter mouth. Size fraction of zooplankton fed to larvae was 100 400 µm.

Results: - Partial supplementation with wild zooplankton increased growth by approx. 2% day -1, and this resulted in fish 4.5-fold larger by Day 60 (see Figures 2 and 3). - Survival in the RA-PH groups was only approx. 20-25% of that measured in the other two groups. Figure 2. Changes in Atlantic cod length and weight when fed different diets during larval rearing.
Results: Representative fish from the 3 groups 60 dph Figure 3. Growth rate of Atlantic cod fed 3 different diets during early development. * indicates a significant difference as compared to the other two groups.
Conclusions and Discussion: Dietary supplementation with wild zooplankton greatly enhanced the growth of Atlantic cod larvae. Growth enhancement seen with partial zooplankton supplementation in this experiment is very similar to that achieved in Norwegian experiments (i.e. approx. 5 fold) in which cod larvae were exclusively fed zooplankton for varying periods (Imsland et al., 2006; Koedijk et al., 2010). Thus, it appears that only a small amount of zooplankton is needed to significantly improve the growth rate of larval cod. Future experiments should be performed to better define the shortest feeding window, and the minimum amount of zooplankton, required to achieve this growth. The protein hydrolysate enrichment used in this experiment did not improve growth, and has a negative effect on survival. The decrease in survival is in contrast to previous research on other marine finfish species including cod. The contrasting results may be due to a difference in the way the protein hydrolysate was processed Freeze vs. Heat Drying?
Future Research: Objective 1: - determine the incidence and type of deformities in 60 dph stained/cleared larvae and in juveniles at the time of PIT- tagging. - monitor growth of the 3 groups for 2 years post - hatch Objective 2: What factor(s) mediated the improved growth in zooplankton fed cod? Nutritional Composition: Diets and larvae. (Lipid and Amino Acid Analyses) Function Genomics: Metagenomics: Gut Histology: Metabolic Physiology: Gene expression related to growth, the stress response, immunology etc. (20K Microarray and QPCR) Characterization of microbial community in the gut. (454 Sequencing) Intestinal morphology / histology. (Electron Microscopy) Basal metabolism, swimming capacity and metabolic scope. (Respirometry) Stress Response: Cortisol levels and gene expression following a combined handling confinement stress (ELISA and QPCR).
