System for stabil ekspresjon av lentivirale strukturelle og regulatoriske proteiner som består av: i. en hybridvektor som omfatter bakuloviral ryggrad som inneholder en integreringskassett flankert av AAV ITRer som inkluderer to ekspresjonskassetter, hvori den første ekspresjonskassetten koder for lentivirale gag og pol gener og den andre for lentiviral rev og en seleksjonsmarkør og ii. et ekspresjonsplasmid som inneholder AAV Rep åpen leseramme (ORF) under kontroll av en promoter. 2. System ifølge krav 1 hvori de to ekspresjonskassettene av hybridvektoren er orientert hale mot hale og hver er drevet av en konstitutiv promoter og bærer en poly A. 3. System ifølge krav 1 eller 2 hvori promoteren er valgt fra CMV, CMV IE, PGK, SV40, ef1α, SFFV og RSV. 4. System ifølge ett av kravene 1 til 3 hvori promoteren er en CMV IE promoter.. System ifølge ett av de foregående kravene hvori seleksjonsmarkøren er valgt fra hygromycin, kanamycin, neomycin, zeomycinresistensgen. 6. System ifølge krav hvori seleksjonsmarkøren er hygromycinresistensgen.

Semi-stabil lentiviral pakkecellelinje som består av celler som stabilt uttrykker lentiviral gag, pol og rev karakterisert ved at slike celler inneholder stabilt integrert til dens genom minst én kopi av en integreringskassett flankert av AAV ITR inkludert to ekspresjonskassetter, hvori den første ekspresjonskassetten koder for lentivirale gag og pol gener og den andre for lentiviral rev og en seleksjonsmarkør.. Semi-stabil lentiviral pakke cellelinje ifølge krav 9 hvori cellen er en human cellelinje valgt fra HEK293, HEK293-T, HEK293-SF, TE671, HT80 eller HeLa. 11. Semi-stabil lentiviral pakkecellelinje ifølge ett av krav 9 eller hvori de to ekspresjonskassettene er orientert hale mot hale og hver ene er drevet av en konstitutiv promoter og bærer en poly A. 12. Semi-stabil pakke cellelinje ifølge ett av kravene 9 eller 11 hvori promoteren er valgt fra CMV, CMV IE, PGK, SV40, ef1α, SFFV og RSV. 13. Semi-stabil pakkecellelinje ifølge krav 12 hvori promoteren er en CMV IE promoter.

3 14. Semi-stabil pakkecellelinje ifølge ett av krav 9 til 13 hvori seleksjonsmarkøren er valgt fra hygromycin, kanamycin, neomycin, zeomycinresistensgen.. Semi-stabil pakkecellelinje ifølge krav 14 hvori seleksjonsmarkøren er hygromycin- resistensgen. 16. Semi-stabil pakkecellelinje ifølge ett av krav 9 til hvori seleksjonsmarkør er klonet nedstrøms for et internt ribosom inngangssted (IRES). 17. Fremgangsmåte for fremstilling av lentivirale vektorer som omfatter å: i. dyrke en semi-stabil pakke cellelinje ifølge ett av kravene 9 til 16, ii. sette inn i den semi-stabile pakke cellelinjen et env gen ved anvendelse av en ekspresjonsvektor, iii. sette inn i den semi-stabile pakke cellelinjen en transfervektor. 18. Fremgangsmåte ifølge krav 17 hvori env genet er valgt fra VSV-G env, MLV 4070 env, RD114 env, kimært kappeprotein RD114-TR, kimært kappeprotein RD114-pro, bakulovirus GP64 env, eller GALV env, eller derivater derav. 19. Fremgangsmåte ifølge krav 17 eller 18 hvori env gen er genet som koder for det kimære kappeproteinet RD114-TR.
