4 1 Patentkrav EP Fremgangsmåte for fremstilling av et modifisert allergifremkallende polypeptid, der fremgangsmåten omfatter trinnene å: (a) modifisere nukleinsyresekvensen som koder for et allergifremkallende polypeptid omfattende en plan overflate, slik at i polypeptidet uttrykt fra den modifiserte nukleinsyren blir strukturen av den plane overflate av nevnte polypeptid endret; (b) uttrykke eller fremstille det modifiserte allergifremkallende polypeptider fra den modifiserte nukleinsyre; og (c) velge de modifiserte allergifremkallende polypeptidene mot hvilke et IgEantistoff spesifikt til nevnte allergifremkallende polypeptid viser minst ti ganger lavere affinitet; karakterisert ved at nevnte plane flate har et areal på Å 2 og bestemmes ved matematisk analyse til å være som den allergifremkallende epitop av β-laktoglobulin definert ved strukturen eller 3D-koordinatene for β-laktoglobulin (SEQ ID NO: 8)- aminosyrene Val43-Lys47 og Leu57-Gln59 i et antistoff-β-laktoglobulinimmunkompleks i henhold til tabell VIII. 2. Fremgangsmåte ifølge krav 1, hvor nevnte polypeptid er valgt fra gruppen bestående av: a) Bos d 5, Beta-laktoglobulin; b) Equ c 1, hestehud; c) Bet v 1, bjørkepollen; d) Bos d 2, flass fra storfe; e) Cyp c 1, karpe-parvalbumin; og f) Hev b 6, lateks. 3. Fremgangsmåte for fremstilling av et modifisert allergifremkallende polypeptid, der fremgangsmåten omfatter trinnene å: a) detektere ved strukturell analyse eller molekylære modelleringsteknikker et potensielt IgE-bindingssete i et allergifremkallende polypeptid;

5 (b) bestemme ved matematisk analyse av en tre-dimensjonal struktur av nevnte allergifremkallende polypeptid om nevnte potensielle IgE-bindingssete inneholder et plant eller flatt overflateareal på Å 2 som den plane flate definert ved 3D-koordinatene til β-laktoglobulin (SEQ ID NO: 8)-aminosyrene Val43-Lys47 og Leu57-Gln59 i et antistoff-β-laktoglobulin-immunkompleks i henhold til tabell VIII, og dersom en plan eller flat overflate som den i β- laktoglobulin blir funnet, fortsetter fremgangsmåten ved å (c) modifisere en nukleinsyresekvens som koder for nevnte polypeptid, slik at polypeptidet uttrykt fra den modifiserte nukleinsyrestrukturen av den plane eller flate overflaten av nevnte polypeptid blir endret; (d) uttrykke eller fremstille et modifisert allergifremkallende polypeptid fra den modifiserte nukleinsyren oppnådd i trinn c).
